The Plums were without a fixture on Saturday, so have managed two training sessions since beating Lydney Town 7 - 0 at the Recruit 12 Community Stadium.
Meanwhile, Tuffley Rovers travelled to Bitton AFC on Saturday in the FA Vase and lost 3 - 1, a result that disappointed manager Neil Mustoe after taking the lead in the game.
Glevum Park will play host to tonight's fixture, a venue where Tuffley will undoubtedly back themselves having only lost twice at home in the league in 2024.
Mustoe has managed Tuffley Rovers since 2021, taking over in the December. After coming through the ranks at Manchester United as a player, the Gloucester born midfielder went on to play for Cambridge United and Yeovil Town whilst featuring heavily for Gloucester City too.
Rovers are currently sat 10th in the Hellenic League and they come into tonight's fixture having won their last three league games; beating Hereford Pegasus and Royal Wootton Bassett Town 1 - 0 after beating Thornbury Town 3 - 1 away from home.
Karnell Chambers will be the one to keep quiet this evening for Janes' side, the former Evesham United and Merthyr Town striker has eight goals in thirteen games for Tuffley.
Brad Bevon backs up Karnell with three goals this season whilst Connor Walters has scored twice.
Macaulay Herbert has just played his 300th game for the Football Club, a mainstay for the club for a number of years now and one that will undoubtedly feature this evening.
Kieran Alder will be another to watch for Rovers this evening, he scored ten goals last season but is yet to get on the scoresheet this campaign. The midfielder, once on the books of West Brom and Cheltenham Town, has Step 4 experience with Cinderford Town and will be imperative to Tuffley's performance tonight.
For Janes' Pershore Town, there could be a welcome return to action for Petar Tonchev who trained on Saturday after returning from Bulgaria.
The Plums will still be without Nathan Benson and Jordan Jones however who are both recovering from injury, with Benson the closer to a return out the two.
Jack Cresswell remains available for selection after his red card against Brimscombe & Thrupp was rescinded following an appeal.
They'll be heavy impetus on Aidan Clark and Bailey Fuller once again for the goals, Fuller has six whilst Clark has five to his name. Harlie Price, James Baldwin, Kirk Layton and Sacha Everard have all scored twice for Pershore this season.
Janes will look to Jude Franks and his defenders once again this evening, they've kept back-to-back clean sheets something Pershore Town haven't done since the end of the 2022/23 Season.
The Plums will be hoping to turn the tide of recent games between the two sides, Tuffley doing the double over Pershore Town last season.
Sixteen teams featuring this evening in Hellenic League Premier Division action, a win for Danny Janes' side would be much welcomed but it's certainly going to be tough.
"An unbelievably tough game, Tuffley’s home record is outstanding and they have secured some great wins already this season." said Janes.
"We come into the game on a good run of form and I expect a performance, we are going there to win.
"To do that, we need every player at it, we have to start fast and maximise moments within the game."
